About The Position

The Research Assistant will work in the Kulkarni laboratory at Joslin Diabetes Center, which studies the biology and function of pancreatic islets, including insulin-secreting beta cells and glucagon-secreting alpha cells, and adult tissue-specific stem cells that form pancreatic islet cells. The position supports flow cytometry and stem cell research, laboratory cell and tissue culture, animal and human tissue studies, and the design, execution, and interpretation of experiments. The Research Assistant also supports laboratory operations by maintaining equipment and records, preparing reagents, ordering and storing supplies, scheduling procedures, and assisting and advising other lab members.

Responsibilities

  • Participate in flow cytometry and stem cell research, operating high-speed, multi-parameter cell sorters and analyzers.
  • Assist Fellows with culturing, maintaining, and banking human tissue cell lines, including skin and blood cells.
  • Assist with mouse tissue cultures, maintenance, and banking of cell lines.
  • Culture pluripotent stem cells, including picking colonies, expanding and freezing cells, and generating mouse embryonic feeder cells.
  • Characterize pluripotent stem cells using teratoma assays, immunohistochemistry, qPCR, and western blotting.
  • Screen offspring of mouse strains by preparing genomic DNA and performing PCR, or by isolating blood cells and using flow cytometry.
  • Place orders for reagents and supplies, log deliveries, and store supplies appropriately.
  • Maintain laboratory plasmid, antibody, and animal databases.
  • Assist with isolating cells and tissues from experimental animals and preparing them for flow cytometry or immunohistochemistry.
  • Assist with isolating cells from human tissue samples and preparing them for flow cytometry or immunohistochemistry.
  • Assist with mouse experimental procedures, including cell transplantation, injection of soluble factors, and routine health monitoring.
  • Work with researchers on experimental design, execution, and interpretation, and assist in training and advising lab members in equipment use.
  • Operate and maintain equipment, schedule procedures, work with multiple investigators, interface with vendors, and prepare reagents.
