Revolution Medicines is a late-stage clinical oncology company dedicated to developing novel targeted therapies for patients with RAS-addicted cancers, with a pipeline of RAS(ON) inhibitors currently in clinical development. The Biophysics and Protein Sciences Group supports drug discovery by generating target proteins using heterologous overexpression systems (bacterial and insect cell expression) and various purification approaches (e.g., bulk purification, FPLC, immunoprecipitation). The company is developing a small-scale expression and purification (SmScExpPur) platform to maximize capacity for testing construct designs and rapidly identifying reliably producible proteins. The summer intern will contribute to this effort by conducting experiments to further develop and evaluate SmScExpPur protocols, aiming to enhance protein yield, consistency, and quality. This will involve executing small-scale expression and purification experiments, analyzing results across different constructs and conditions, and providing data-driven recommendations for protocol improvements. The proteins generated will be used for basic quality control assessments to determine protein identity, aggregation state, and folding.
