About The Position

Join MilliporeSigma as a strategic R&D team lead within Chemical Synthesis Solutions (CSS) R&D. This pivotal role serves as the scientific bridge between chemistry and biology, requiring a dual-domain expert: a chemist with deep protein/peptide/DEL and medicinal chemistry foundations who can independently design and execute in vitro biochemical and cell-based assays to validate chemical hypotheses. The work spans the full early drug discovery continuum, from target identification and mechanism of action (MOA) studies, through proof-of-concept (PoC) validation of CSS chemical products and proprietary platforms, to enabling the characterization of novel therapeutic modalities including peptide drugs, ADCs, ASOs, and targeted protein degraders. The role also involves spearheading the design, construction, and strategic application of DNA-Encoded Libraries (DELs) for high-throughput affinity screens. You will lead a cross-disciplinary sub-team and act as a senior scientific partner to customers, Key Opinion Leaders, and internal stakeholders, ensuring CSS remains the partner of choice for novel therapeutic modalities. Additionally, you will sustain strategic momentum across adjacent domains like next-generation biomaterials and drug delivery, and shape the 5–10 year chemical biology capability roadmap for CSS. The ideal candidate is a chemist at heart with a strong biological toolkit, understanding how molecular structure drives biological function and translating chemical innovation into biologically validated insights for product and platform strategy.

Requirements

  • PhD in Chemical Biology, Medicinal Chemistry, Bioorganic Chemistry, Biochemistry, or a closely related discipline with a strong chemical biology component.
  • Minimum 10 years of post-doctoral R&D experience in pharma, biotech, or life science tools (industry or academic), with a demonstrated track record spanning both chemistry and in vitro biology.
  • Deep, hands-on expertise in chemical biology: chemical probe design and synthesis, ABPP, photoaffinity labeling, chemoproteomics, or equivalent target identification and MOA methodologies.
  • Strong protein and peptide chemistry expertise: SPPS/LPPS, peptide conjugation, cyclic/stapled peptide design, site-specific protein modification, and bioconjugation strategies for ADCs, PROTACs, and peptide therapeutics.
  • Solid medicinal chemistry literacy: SAR, ADME, selectivity optimization, and drug-likeness principles applied to small molecules, peptides, and chemical probes.
  • Deep technical expertise in mammalian cell culture, biochemical and cell-based functional assays, binding assays, and protein labeling — with the ability to independently design, develop, and execute these workflows — and a strong focus on characterizing novel modalities (ASOs, RNA, proteins, ADCs, peptide drugs, PROTACs, etc.).
  • Comprehensive understanding of MOA study design across diverse therapeutic target classes (kinases, proteases, E3 ligases, nucleic acid targets, GPCRs, etc.).
  • Hands-on expertise in the construction and utilization of DNA-Encoded Libraries (DELs-ssDELs, dsDELs), encompassing library design, split-and-pool synthesis, DNA-compatible chemistry, affinity selection screening, and hit deconvolution/sequencing analysis.
  • Proven track record leading R&D teams: sustaining scientific performance, managing cross-domain project portfolios, and driving deliverables from concept to validated output.
  • Strong strategic and analytical skills: able to connect scientific workflows to business outcomes, project milestones, and budget accountability.
  • Excellent communication skills: experienced in presenting complex chemical biology and in vitro data to multidisciplinary audiences including senior leadership, customers, and KOLs.

Nice To Haves

  • Experience with targeted protein degradation (PROTAC/molecular glue) chemistry and associated cellular validation assays (FACS-based degradation, western blot, TR-FRET, NanoBRET).
  • Familiarity with nucleic acid-based modalities (ASO, siRNA, mRNA) and their chemical modification strategies (phosphorothioate, 2’-OMe, LNA) and functional cellular readout assays.
  • Experience integrating automation, liquid handling, and AI/ML tools into chemical biology or assay workflows to accelerate the DMTL cycle.

  • Track record of successful external collaborations (academic institutions, CROs, industry partners) resulting in co-developed products, publications, or IP.

Responsibilities

  • Design and synthesize chemical probes, tool compounds, and novel building blocks; apply SAR, ADME, and selectivity principles to guide prioritization of CSS chemical candidates and platform molecules.
  • Lead protein and peptide chemistry within CSS R&D: solid-phase/liquid-phase peptide synthesis (SPPS/LPPS), cyclic and stapled peptide design, site-specific protein modification, and antibody conjugation strategies for ADCs, PROTACs, ASOs and peptide therapeutics.
  • Lead the design and synthesis of novel DELs (encompassing membrane-DEL and proximity-DEL) and integrate DEL-derived insights to streamline and accelerate validation of screening hits.
  • Provide expert guidance on linker chemistry, bioconjugation, and payload design for novel modalities (ADCs, TPDs, peptide–drug conjugates, ASO-based therapeutics, as well as RNAs).
  • Deploy chemical biology approaches — activity-based protein profiling (ABPP), photoaffinity labeling, chemoproteomics, chemical genetics — to interrogate target biology and support target identification workflows.
  • Independently design, develop, optimize, and execute a comprehensive suite of in vitro biochemical and cell-based assays: binding assays (TR-FRET, AlphaScreen, SPR, fluorescence polarization), enzyme activity assays, cellular target engagement assays (CETSA, NanoBRET), and functional/phenotypic assays.
  • Apply protein labeling techniques — fluorescent labeling, biotinylation, crosslinking, affinity enrichment, pull-down, and co-immunoprecipitation — to support target deconvolution and MOA studies.
  • Maintain mammalian cell culture capabilities required for functional assay execution and PoC validation of CSS chemical candidates and platforms.
  • Provide deep technical expertise in the in vitro characterization of novel therapeutic modalities: peptide drugs, ADCs, ASOs, siRNA/mRNA constructs, proteins, and targeted protein degraders, including assessment of cellular uptake, target engagement, and functional activity.
  • Establish and enforce assay quality standards (Z’-factor, S/N ratio, CV, reproducibility) to ensure data integrity and scientific rigor across all biological testing activities.
  • Serve as the primary scientific resource for MOA studies and PoC validation across CSS chemical candidates and proprietary platforms; define experimental strategies, execute studies, and deliver clear, decision-enabling data packages with explicit go/no-go criteria.
  • Lead target identification and target engagement experiments using chemical biology (ABPP, photoaffinity labeling, chemoproteomics) and biochemical approaches (binding assays, CETSA, NanoBRET).
  • Validate proprietary CSS building blocks, linker platforms, late-stage functionalization (LSF) reagents, and screening libraries through relevant biological assays; translate findings into scientific narratives that support product positioning and IP strategy.
  • Define and execute a 5–10 year chemical biology and in vitro biology capability roadmap for CSS, aligned to the Innovation Strategy 2030 and LS Innovation Vectors (Novel Modalities, Next-Generation Biology).
  • Expand CSS capabilities in novel therapeutic modalities — peptide drugs, ADCs, ASOs, RNA therapies, targeted protein degraders — identifying and closing capability gaps that limit customer workflow support.
  • Monitor the competitive landscape, scientific literature, and patent environment; identify breakthrough innovation opportunities and translate them into actionable R&D investment proposals.
  • Integrate automation, miniaturization, and AI/ML tools into chemical biology and assay workflows to accelerate the Design-Make-Test-Learn (DMTL) cycle.
  • Lead and develop a cross-disciplinary sub-team spanning chemical biology, biochemistry, and protein/peptide chemistry; provide scientific direction, mentorship, and performance management.
  • Sustain strategic momentum across adjacent domains including next-generation biomaterials, 3D bioprinting, and drug delivery; apply TRL/CRL frameworks to assess project maturity and protect delivery timelines against stage-gate criteria.
  • Own the sub-team budget (CAPEX and OPEX): plan, forecast, track, and control spend; develop compelling business cases (NPV, TAM/SAM/SOM) for new capability investments.
  • Build and maintain a collaborative network across CSS R&D, Medicinal Chemistry, Materials Science, Engineering & Digital, and Business Development, as well as external partners (CROs, CDMOs, academic institutions).
  • Engage customers, Key Opinion Leaders, and scientific advisory boards to translate market needs into actionable R&D roadmaps; represent CSS at scientific conferences and customer-facing engagements.
  • Identify patentable inventions arising from chemical biology and protein/peptide work; manage IP disclosures and collaborate with legal teams on freedom-to-operate assessments.
  • Champion sustainability and green chemistry practices within chemical biology workflows; foster a culture of continuous learning and scientific excellence across the team.
