About The Position

The Stanton Laboratory in the Center for Genomic Medicine (CGM) at Massachusetts General Hospital (MGH) is seeking a motivated, enthusiastic candidate to join our research group. Our research group works with brain 3D organoid model to understand the biological complexity, target identification, and validation in neurodegenerative disease. We are seeking a highly motivated Research Technician to join our research team focused on human induced pluripotent stem-cell (iPSC) models, gene editing, cellular phenotyping, and 3D brain organoid systems. The successful candidate will play a central role in supporting projects involving iPSC culture, CRISPR-based genome editing, directed differentiation, molecular cloning, and downstream validation assays. This position is ideal for someone with strong hands-on experience in mammalian cell culture and molecular biology who is excited to contribute to a fast-paced, collaborative research environment. The technician will be expected to maintain rigorous experimental records, generate reproducible data, and assist in day-to-day laboratory operations.

Requirements

  • Bachelor's Degree in a Related Field of Study required.
  • Prior laboratory experience, either through academic coursework, internships, or employment (1-2 years preferred).
  • Strong theoretical and practical knowledge of laboratory techniques and procedures.
  • Familiarity with common laboratory equipment and instruments used in research experiments.
  • Proficiency in data collection, analysis, and scientific software tools.
  • Excellent attention to detail, organizational skills, and the ability to multitask and prioritize tasks effectively.
  • Strong problem-solving skills and the ability to troubleshoot technical issues.
  • Excellent written and verbal communication skills to effectively convey experimental procedures, results, and challenges.

Nice To Haves

  • Prior experience with human iPSC culture

Responsibilities

  • Perform laboratory experiments following established protocols and procedures.
  • Prepare and handle samples, reagents, and solutions, ensuring accuracy and adherence to safety guidelines.
  • Operate and maintain laboratory equipment and instruments, conducting routine calibration and troubleshooting as needed.
  • Record experimental data accurately and maintain organized records of procedures, protocols, and results.
  • Compile and analyze data using appropriate statistical and analytical methods.
  • Assist in data interpretation and presentation, contributing to research findings and reports.
  • Maintain and expand human iPSC lines under sterile culture conditions.
  • Conduct CRISPR-based gene editing workflows in iPSCs, including transfection/nucleofection, lentiviral transduction, clone handling, and expansion.
  • Support generation, validation, and maintenance of edited cell lines.
  • Perform directed differentiation of iPSCs into relevant downstream cell types.
  • Generate, culture, and maintain 3D brain organoids.
  • Assist with optimization and routine handling of organoid-based workflows.
  • Validate differentiated cells and organoid-derived cultures using Flow cytometry, Immunofluorescence staining and imaging, and perform functional assay as required.
  • Carry out molecular cloning tasks such as plasmid preparation, bacterial transformation, restriction digest analysis, Gibson assembly, PCR, and construct verification.
  • Assist with genotyping, sequencing library preparation, and clone validation.
  • Maintain accurate, organized, and detailed lab notebooks and experimental records.
  • Contribute to protocol optimization, troubleshooting, and experimental planning.
  • Present data clearly in lab meetings and communicate progress to the research team.
