About The Position

The Research Department at Alnylam is seeking a Principal Scientist to join a growing team responsible for advancing RNAi therapeutics into the future, by opening up targeted delivery to all tissues. The successful candidate will be a key leader in our ligand discovery efforts, responsible for driving end-to-end ligand discovery projects for novel receptors. This role requires a deep understanding of antigen design, antibody engineering, immunology, and high-throughput screening technologies. This position is onsite and will be primarily located in Cambridge, MA.

Requirements

  • PhD in Molecular Biology, Bioengineering, Biochemistry, Cell Biology, or a related field.
  • A minimum of 6 years of post-PhD experience in the biopharmaceutical industry, with a primary focus on antibody discovery and engineering.
  • Proven track record of successfully leading antibody discovery programs from inception to lead candidate delivery.
  • Hands-on experience with single B cell isolation and screening technologies is required; direct experience with the Bruker Beacon platform is required.
  • Deep expertise in molecular biology techniques for antibody cloning, expression, and sequence analysis, including extensive experience with NGS library preparation and data interpretation.
  • Demonstrated experience developing and running high-throughput, automated screening assays.
  • Experience with managing external research collaborations and CROs for immunization and discovery campaigns.

Responsibilities

  • Lead the design and implement innovative immunization strategies, including the identification, selection, and management of external immunization vendors (CROs).
  • Establish and oversee high-throughput B cell screening and selection workflows.
  • Develop and integrate a robust Next-Generation Sequencing (NGS) workflow for the rapid identification and analysis of antibody variable region sequences from screening campaigns.
  • Drive the design, execution, and automation of high-throughput screening cascades to characterize antibody properties such as affinity, specificity, and function.
  • Engineer complex antibody modalities, including Mult specific antibodies, nanobodies, and evaluate novel protein formats.
  • Collaborate with cross-functional teams, including protein science, pharmacology, and translational science to ensure project goals are achieved.
  • Manage project timelines, resources, and data analysis, and present findings to internal teams and senior leadership.
