About The Position

The California Department of Public Health’s Microbial Diseases Laboratory Branch is responsible for diagnostic and surveillance testing, applied research projects, and education and training for the identification and characterization of bacterial, mycobacterial, fungal, and parasitic pathogens of public health importance. Under the direction of the Supervisor of the Mycobacterial and Parasitic Diseases Reference Unit, within the Mycobacterial, Mycotic & Parasitic Diseases Section, the Microbiologist II performs duties associated with sample receipt, processing, characterization, and detection of Mycobacterium tuberculosis complex (MTBC) pathogens. The Microbiologist II will be involved in a research study on early MTBC detection from patients using innovative and complex approaches and methods. The Microbiologist II will assist with developing and validating an efficient, scalable, and operationally practical workflows for extraction of MTBC DNA from various matrices. Some functions performed by the Microbiologist II will be conducted under Biosafety Level 3 conditions, requiring the use of appropriate respiratory protection and following requested occupational health screenings.

Requirements

• Ability to perform micromanipulations such as pipetting small volumes and other fine measurements using laboratory instrumentation and equipment.
• Knowledge of standard biosafety precautions for working with potential infectious material, sterile technique, molecular contamination control, understating of molecular methods (e.g., RT-PCR) and DNA extraction principles.
• Bachelor’s degree in microbiology or related biological science from an accredited university.
• Minimum of two years of experience handling infectious agents in a microbiology laboratory setting; preferably in a clinical or public health laboratory settings performing high complexity testing.
• Minimum of two years of experience with molecular techniques, like RT-PCR, sequencing, etc..
• Computer skills (Microsoft Word, Excel, Outlook, PowerPoint, SharePoint)
• Ability to effectively communicate verbally and in writing.
• Ability to work independently and adapt to varied work settings.
• Ability to work effectively with a team of diverse individuals.
• Ability to work in BSL-3 conditions with required respiratory protection

Responsibilities

• Receives human specimens and performs data entry into the Laboratory Information Management System (LIMS) or other databases.
• Performs DNA extraction and purification in accordance with an approved protocol.
• Performs MTBC detection in human specimens using real-time Polymerase Chain Reaction (RT-PCR)
• Assess the efficacy of MTBC inactivation under the test conditions to ensure safety of the material for operation.
• Develops, optimizes, and validates efficient and scalable method(s) for extracting and amplifying MTBC DNA from human specimens.
• Performs molecular and phenotypic microbiological techniques applicable to MTBC detection and characterization, e.g. DNA extraction, PCR, culture, microscopy, staining.
• Develops and optimizes workflows, protocols, flowcharts, and standard operating procedures (SOPs) for research study intended for internal use and sharing with external partners.
• Reviews test results for completeness and accuracy.
• Maintains accurate, neat, and thorough laboratory documentation. Ensures appropriate entry of data and reporting of results to the study investigators.
• Performs all work in compliance with MDL, Section, and Unit QC/QA policies for laboratory operations including biosafety, chemical hygiene, quality, and other applicable standards.
• Performs packaging and shipping of samples on a regular basis.
• Communicates effectively with internal and external partners. Coordinates sample shipment and receipt with external partners.
• Manages and maintains storage of biological materials, ensures appropriate storage conditions and inventory.
• Maintains inventory of essential chemicals, reagents, and disposables. Assists with ordering of supplies and order tracking.
• Prepares and performs QC of reagents.
• Compiles data and prepares test reports and other technical reports related to grants and projects. Assists with preparation of manuscripts and presentations. Participates in calls and in-person meetings.
• Performs other related laboratory and data management duties as assigned.
