About The Position

The University of Pittsburgh School of Medicine seeks a Laboratory Research Technician IV to work in Pittsburgh, PA. Duties include designing/performing: (i) isolation, activation, and expansion of T cells (ii) cryopreservation, sub-culturing, and revival of various cell lines and primary cells; (iii) cell-based assays including cytotoxicity, cell viability, and cell proliferation assays; (iv) immunofluorescence imaging using confocal microscopy and single-cell time-lapse imaging; (v) isolation and handling of PBMCs from human whole blood; (vi) multicolor flow cytometry, SDS-PAGE, western blot, multi-mode reader-based assays and various biochemical assays; and (vii) data analysis using Graphpad Prism, FlowJo, Excel and Image-J.

Requirements

  • Master’s degree (or foreign equivalent degree) in Biology, Biotechnology, or a related field
  • One (1) year of experience in a related position
  • Experience working with Cryopreservation, sub-culturing and revival of various cell lines, mammalian cell lines and primary cells
  • Experience with In-vitro assays such as cytotoxicity, cell viability, cell proliferation
  • Experience with Live cell time-lapse imaging for cell viability assays, spheroids, immune cell killing, co-culture assays and image analysis
  • Experience with Isolation of and assays using Peripheral Blood Mononuclear Cells (PBMCs) from human blood
  • Experience with isolation and expansion of naïve T cells, CD4 T cells, CD8 T cells, and regulatory T cells
  • Experience with Activation and expansion of T cells using Dynabeads and plate coated antigen
  • Experience with Screening of peptide pools, cytokine/chemokine profiling using LUMINEX, and direct, indirect, Quantikine, and sandwich ELISA assays
  • Experience with Seed, treat, fix, permeabilize, block and antibody staining/addition of cell lines and primary immune cells for multicolor flow cytometry of surface and intracellular markers using appropriate compensation and FMO controls for different multi-point in-vitro assays
  • Experience with Analysis of the flow cytometry data for receptor quantification and drug screening using Flow-Jo Software
  • Experience with Preparing cells for molecular biology techniques such as SDS-PAGE, western Blot, protein/antibody quantification using Bradford/BCA assay and other multi-mode reader-based assays, bacterial transformation and cloning
  • Experience with data analysis using Graphpad Prism and Excel
  • Experience with Image analysis using Image-J
