About The Position

Century Therapeutics is seeking a highly motivated Sr. Research Associate or Associate Scientist focused on targeted gene editing of iPSCs for pipeline programs and providing molecular assay support across functional groups within Research. This role requires a strong understanding of CRISPR-Cas9 based editing as well as proficiency in designing and conducting molecular assays. The ideal candidate brings a strong collaborative mindset and works closely with other functional groups.

Requirements

• Senior Research Associate: BS or MS in biology or related field with ~2-3+ years of relevant experience.
• Associate Scientist: BS or MS in biology or related field with ~3-5+ years of relevant experience.
• Extensive cell culture experience is essential, experience with IPSC culture highly preferred.
• Hands-on experience using CRISPR-Cas9 preferably in iPSCs
• Experience with multi-color flow cytometry is essential. Cell sorting experience is a plus. Familiarity with FlowJo a plus.
• Proficiency in molecular biology techniques including molecular cloning, qRT-PCR and ddPCR is required.
• Excellent verbal and written communication skills with demonstrated ability to present findings to a broad audience.

Responsibilities

• Gene editing of iPSCs using CRISPR-Cas9 under GxP principles.
• Design, develop and validate assays to confirm the desired editing in iPSC clones.
• Design and optimize guide RNAs (gRNAs) targeting specific regions within the genome and validate results using NGS.
• Utilize molecular cloning skills to generate donor vectors for Homology Directed Repair (HDR)
• Conduct qRT-PCR for gene expression and ddPCR assays for establishing transgene copy numbers in edited cells in accordance with established protocols and guidelines.
• Lentivirus transduction of iPSCs and target cell lines optimizing MOI for high efficiency.
• Analyze and document experimental data using electronic lab notebooks.
• Author, review and update SOPs, batch records and other relevant documentation.
