About The Position

PTC Therapeutics is seeking an innovative and motivated scientist to join our Multiomics team in Mountain View, CA. The successful candidate will develop and apply chemoproteomic methods to advance drug discovery programs and translational biomarker strategies. We are looking for someone to join our group who is: organized, methodical, team-oriented, and not afraid to tackle challenging problems. The ideal candidate will be a technical expert in proteomics and have broad knowledge of biological processes associated with either: neurodegenerative/neuromuscular disorders, inflammation, immunology, or ferroptosis. The Scientist I is responsible for the detailed planning and execution of scientific experiments under the guidance of the Executive Director of Multiomics. The incumbent works in matrixed teams and serves as a subject matter expert for programs advancing PTC’s research and drug discovery activities. The Scientist I supports adherence to relevant regulatory requirements and internal Standard Operating Procedures (SOPs) as appropriate.

Requirements

  • Experience in maintaining and troubleshooting nano-LC instruments as well as ion-mobility or high-resolution mass spectrometers.
  • Extensive use of sample preparation and enrichment strategies for chemoproteomics, global proteomics, and PTM analysis.
  • Expertise in using proteomic software tools for ID, statistics, pathway analysis, and visualization.
  • Detail-oriented and organized with excellent problem-solving skills.
  • Self-motivated and also a team player.
  • Proactive communicator with a positive attitude.
  • Ability to multitask and respond quickly to shifting priorities while adhering to program timelines.
  • Ph.D. in Analytical Chemistry, Chemical Biology, Biochemistry, or Biomedical Engineering with 4 – 5 years of relevant experience (including doctoral…).

Nice To Haves

  • Prior use of Mass Dynamics software.
  • Coding experience is a plus.
  • Hands-on use of Bruker tims-ToF instruments running DIA-PASEF, PRM-PASEF, and diagonal-PASEF.
  • Previous use of Evosep systems run at ≥ 100spd or prior use of the Bruker nano-Elute LC.
  • Expertise using proteomic software tools such as: Spectronaut, MS Fragger, DIA-NN, etc.

Responsibilities

  • Sample processing and enrichment of multiple sample types ( e.g. plasma, CSF, tissues, and cells) for downstream proteomics analysis using a variety of techniques including, but not limited to: ultracentrifugation, SP3, SAX, RP/C18, IMAC/MOAC, SEC, FASP, IP, co-IP, etc.
  • Daily operation of nano-LC ( i.e. Evosep and nano-Elute) instruments as well as ion-mobility and high-resolution mass spectrometers ( i.e. Bruker tims-ToF and Thermo Orbitrap).
  • Use of label-free chemoproteomic techniques like LiP and TPP as well as directed proteomic analyses such as ABPP or PAL.
  • Execution of quantitative proteomic workflows using TMT, SILAC, spiked-in isotopically labeled standards, or PRM analysis.
  • Analysis of proteomic datasets using Spectronaut, MS Fragger, DIA-NN, Skyline, etc.
  • Integrates multiomic datasets and uses pathway analyses to correlate observed changes with disease phenotype, treatment effect, or translational biomarker identification.
  • Communicates effectively and cooperates with cross-functional teams to plan and execute strategies to achieve program and corporate goals.
  • Tracks daily progress in Agile software to provide real-time updates for project stakeholders.
  • Represents the organization in external collaborations and at scientific conferences.
  • May participate in the authorship and publication of scientific papers and patents.
  • Interviews and evaluates potential new hires.
  • Acts as a resource for and trains junior staff.
  • Abides by all safety and regulatory guidelines.
  • Maintains a detailed laboratory notebook.
  • Contributes to general lab needs such as: equipment maintenance, evaluation of capital equipment purchases, and authorship/development of Standard Operating Procedures (SOPs).
  • Performs other tasks and assignments as needed and specified by management.
