The Alloy Genetic Medicines group is innovating a novel RNA-based drug discovery platform, namely “AntiClastic”, spanning the fields of antisense therapeutics to immunomodulating therapeutics. The team works closely with external partners to discover and develop AntiClastic RNA medicines. As a Research Associate / Associate Scientist, you will work within the Genetic Medicine team in Waltham, MA. You will also collaborate with internal/external partners on nucleic acid drug discovery projects to ensure timely execution of the experiments, and delivery of high-quality data. This role will have an autonomy owning various stakeholders, projects, and tasks in a dynamic start-up environment while keeping up the pace of Alloy.

Requirements

  • B.S. or M.S. degree in Biology or related science.
  • 1-3 years of additional experience in biotech/pharma (Research Associate), 4-6 years of experience (Principal Associate Scientist).
  • Knowledge of antisense oligonucleotide modalities and their mechanism of action.
  • Experience designing and executing molecular biology, protein and cell-based assays.

Nice To Haves

  • Experience with animal handling (e.g. dissection, surgery, injection, imaging)
  • Working experience with oligonucleotide and/or siRNA
  • Experience of design and execution of Bioanalytical assays (e.g. MSD, ELISA)

Responsibilities

  • Design and execute several molecular biology and protein techniques including, but not limited to, nucleic acid isolation, RT-qPCR (ddPCR), Western blots and ELISA.
  • Culture various cell types including established cell lines, primary cells and iPSCs.
  • Perform a range of cell-based assays, including those utilizing high content imaging systems and plate readers for bright field/fluorescence/luminescence detection.
  • Process rodent tissue for RNA extraction and downstream analysis to assess target RNA and ASO levels.
  • Run various assays to assess ASO secondary structure formation, inter and intra-molecular ASO interaction and ASO-protein interaction.
  • Operate high-throughput tools, including 96 channel electronic pipets to streamline and enhance experimental workflows.
  • Analyze data using GraphPad software and prepare presentation-ready images to effectively communicate experimental findings.
  • Maintain comprehensive and accurate records of all experiments using the Benchling electronic notebook, ensuring meticulous documentation and data integrity.
